摘要
目的:考察流动相对高效凝胶渗透色谱法(HPGPC)测定灰树花多糖(GFP)相对分子质量(Mr)的影响因素。方法:采用HPGPC法分别以不同的流动相、盐浓度、pH值测定GFP的Mr,并对测定结果进行分析。结果:分别以纯水、0.02%NaN3、0.1mol·L-1NaNO3和0.1mol·L-1Na2SO4溶液为流动相进行测定,GFP的Mr依次为822410、177520、29112和25066道尔顿;在0-0.025mol/L氯化钠溶液中Mr先急剧降低,然后随着盐浓度的增加,在0.1-0.2mol/L氯化钠溶液中趋于平稳;在pH3-6时,Mr随pH增加而急剧增加,在pH6-9范围内变化较小,当pH为10时又稍微增加。结论:不同流动相、盐浓度、pH值对GFP的Mr测定结果有明显影响。
Objective:To investigate the mobile phases influence on relative molecular weight(Mr) determination of Grifola Frondosa Polyaccharide(GFP) by High performance gel permeation chromatography(HPGPC). Methods: Determine Mr of GFP by HPGPC using different mobile phases, concentrations of NaC1 and pH value of GFP separately. Results:The Mr of GFP are 822410,177520,29112 and 25066 dalton determined by HPGPC using pure water,0. 02%NAN3 solufion,0.1mol·L-1 NaNO3 solution and 0. 1mol·L-1 Na2SO4 solution as mobile phase respectively. The Mr of GFP was sharply dropped under the eoneentration of 0 - 0. 025 mol/L NaCl and then tend to be stable from 0. 1 to 0. 2 mol/L NaC1. The Mr increase quickly from pH 3 to 6 ,and then maintain stable from pH 6 to 9 ,but slightly inereased at pH 10. Conclusion:Different mobile phases, concentrations of NaCl and pH value have great influence on Mr deterroination of GFP.
