互为内标法测定奈替米星和链霉素的血药浓度

摘要 目的:建立人血清中硫酸奈替米星和硫酸链霉素的浓度的HPLC测定方法。方法:采用色谱柱KromnsilC18柱(250mm×4.6mm,5μm),流动相为乙腈-磷酸盐缓冲液(10:90,pH2.5,含10mmol/L庚烷基硫酸钠),流速0.8mL/min,柱温40℃,检测波长205nm。奈替米星和链霉素互为内标。结果:本法奈替米星和链霉素的线性范围分别为:2mg·L-1~20mg·L-1(r=0.9995,n=6)及1mg·L-1~60mg·L-1(r=0.9992,n=6)。回收率分别为100.9%及99.8%,奈替米星的日内日间RSD分别为3.95%、5.10%;链霉素的日内日间RSD分别为4.22%、5.09%。结论:本法灵敏度较高,结果准确可靠,操作简便,适合临床常规血药浓度监测。 Objective:To establish a RP -tIPLC method for determination the concentration of netilmicin and streptomy- cin in human plasma. Methods:The determination was performed on Kromnsil C 18 colunm under 40℃ ;the mobile phase con- sisted of acetonitrile -phosphate buffer( 10:90, pH 2. 5, containing 10mmol·L-1 sodium heptanesulfonate)with a flow rate at 0. 8mL·L min-I, detecting wavelength at 205nm. Netilmicin and streptomycin were used as internal standard each other. Results: The line range of netilmicin and streptomycin curve were 2 - 20 mg·L L- 1 ( r = 0. 9995, n = 6) and I - 60 mg·L L-1 ( r = 0. 9992, n = 6), The average recovery rate were 100. 9% and 99. 8% respectively, The intra - RSD and inter - RSD of netilmicin were 3.95% and 5. 10%, of streptomycin were 4. 22% and 5.09%. Conclusion:h appears to be a rapid, convenient and accurate method for determination of netilmicin and streptomycin concentration in serum.
