摘要 本试验以高粱雄性不育系和保持系为材料进行金属离子诱导应激反应的研究。结果表明CuSO_4溶液诱导高粱雄性不育系产生的应激反应明显,所用CuSO_4溶液最适浓度为250pmol/L,最适培养时间为4小时。ZnSO_4溶液诱导反应不明显。电泳分析和自显影检测出CuSO_4诱导产生新蛋白区带在3197A为9条,3197B为20条。但各蛋白带产生所需浓度和时间不完全相同,表明各种蛋白的基因表达是相互独立的,且这些基因的表达随发育而有变化。从分析处理后的4种酶的反应可以看出,其中细胞色素氧化酶有5条带,过氧化物酶反应灵敏,但只有2条带。通过CuSO_4处理与热激处理比较,推测3197A、3197B有其特殊的基因调控系统。从产生的可溶性蛋白或酶来看,CuSO_4能使3197A产生的应激蛋白趋向于3197B,说明两者具有相似之处。 On the basis of our study of heat shock proteins of male sterile sorghum induced by heat shock (after our found the phenomenon of male sterile sorghum change to male ferile pheno-type induced by high temperature 40℃), we began a study of the heat shock responsa (it can be name stress response) of male sterile sorghum and its maintainer induce with metal ion in 1987. The exhibited new protein bands after treated by metal ion can be name stress proteins.The 5-day-old seedlings of male sterile sorghum and its maintainer were treated in the medium containing CuSO4, the control were incubated in the medium without CuSO4, and the soluble protein were extracted and analysed with SDS-polyacrylamid gel electrophoresis (SDS-PAGE). The results are as follows:1. The most appropriate incubated concentration of CuSO4 is 250 μmol/L, the appropriate incubated time is 4 hours.2. The CuSO4 treated male sterile sorghum exhibited 9 SPs bands as comparied with its control. They are 58 kd, 52 kd, 46 kd, 41 kd, 38 kd, 28 kd, 18 kd, 16 kd, and 15 kd.The CuSO4 treated maintainer exhibited 20 changed protein bands in which bands were not observed in control, they are 95 kd, 86 kd, 72 kd, 70 kd, 66 kd, 64 kd, 62 kd, 60 kd, 55 kd, 50 kd, 46 kd, 42 kd, 40 kd, 36 kd, 28 kd, 24 kd, 22 kd, 17 kd, 16 kd and 15 kd.3. Studies indicating CuSO4 concentration and incubated time that induced those new proteins in male sterile sorghum are ex'tremely different. The results clearly show that the expresion of stress genes of different type plant are independent.
