摘要
该文介绍用2组寡核苷酸引物X_1,X_2和Y_1,Y_2分别扩增人类X染色体和Y染色体的α类卫星DNA重复序列来鉴定胎儿性别。PCR扩增产生的Y染色体特异DNA片段可进一步用限制酶Dde Ⅰ和Hinf Ⅰ消化来分析鉴定。本方法灵敏而准确,作者应用此法对2例DMD高危胎儿进行了性别鉴定。
This paper describes the determination of fetal sex by PCR amplification of alphoid satellite sequences on human X and Y chromosomes using two sets of oligonucleotide primers X_1 X_2 and Y_1, Y_2. When female DNA was used as a template for the polymerase chain reaction, only the expected 130 bp X chromosomespecific fragment was detected while with male DNA both the expected 170 bp Y chromosome-specific and 130 bp X chromosome-specific fragments were detected. By using this sensitive and accurate method, the sex of the two fetuses at risk for DMD was determined.
