酸性β-1.4葡萄糖苷酶的免疫测定法及其临床应用

摘要 从胎盘中提纯酸性β-1.4葡萄糖苷酶(acid β-1.4-glucosidase,GβA)作为抗原免疫家兔,获得免抗人GβA抗体。根据抗体免疫特性,建立了具有高度特异性的GβA免疫测定法。利用该法对绒毛和脾组织中的β-1.4葡萄糖苷酶进行了检测和分析,证明:(1)绒毛中只含有GβA,不含中性β-1.4葡萄糖苷酶;(2)对照者脾和Gaucher病患者脾中GβA活性分别为51.99±16.08和2.51±0.99nmol·(mg prot·hr)^(-1),后者酶活性仅相当于前者的4.8%,证明免疫测定法对Gaucher病具有可信的诊断价值。 With the purified GβA as an Ag, We prepared rabbit anti-GβA antibody. On the basis of studying immune property of the Ab, a special immunoassay was established for GβA. Then the β-1.4-glucosidase properties and GβA activities from villi, control spleens and Gaucher's spleens were determined by this method. The results show:there is only GβA, no GβN, in villi; the GβA activities in spleen tissues from controls and Gaucher patients are 51.99±16.08 and 2.51±0.99μ mol. (mg prot·hr)^(-1) respectively. The residual activity of GβA in Gaucher spleens is only 4.8% of the normal mean, which proved that this special immune assay is convincingly valuable to clinical diagnosis.
