一种新的氨基葡萄糖苷酶检测底物的合成

摘要 首次合成了N-乙酰-β-D-氨基葡萄糖苷酶检测底物———2-氯-4-乙酰基苯基-N-乙酰-β-D-氨基葡萄糖苷(CAP-NAG),并对其合成方法进行了研究,最终找到一种操作简便,成本较低,收率较高的合成方法。合成方法如下:以2-氯-4-乙酰基苯酚和氯代乙酰氨基葡萄糖为原料,在丙酮-无水碳酸钾体系中进行缩合反应,得到固体粗品经重结晶后,在甲醇-甲醇钠体系中进行脱乙酰基反应即得标题化合物,产物结构经红外光谱、核磁共振谱及元素分析确证。 A novel substrate 2-chloro-4-acetylphenyl-N-actyl- β-D-glucosaminide(CAP-NAG) for determination of N-acetyl-β-D-glucosaminidase was synthesized and the synthesis methods were invesitigatede. A method was established which is convenient in operation, lower in the cost and higher in the yield. Method as follow:the 2-chloro-4-acetylphenol and 3,4, 6-triacetate-2-acetamido-2-deoxy-a-D-glucosyl chloride was used as raw material and the crude product was obtained by condensation reaction in acetone-potassium carbonate system. The target compound was obtained via recrystailisation and deacetylation reaction in methanol-sodium methoxide system. The compound was examined by IR, ^1HNMR and elemental analysis to verify its compliance with the expected structure.
