氨基胍对糖尿病(消渴)大鼠肝脏氧化应激及线粒体CYP450 2E_1的作用 被引量:1

摘要 目的研究氨基胍对糖尿病(消渴)大鼠肝脏氧化应激状态的影响及线粒体CYP450 2E1的干预作用。方法 28只三月龄健康雄性W istar大鼠,随机化分为3组。正常对照组(NC组)8只,其余采用链脲佐菌素腹腔注射建立糖尿病模型。造模后一周将糖尿病大鼠分为糖尿病对照组(DM组),氨基胍治疗组(Ag组)。治疗9周后处死大鼠。采用比色法测定大鼠肝脏匀浆和肝脏线粒体内MDA、Cu/Zn-SOD及T-AOC。采用比色法测定大鼠肝脏线粒体CYP450 2E1酶活力。结果与NC组相比,DM组大鼠肝脏匀浆和肝脏线粒体的MDA含量显著升高(P<0.01),Cu/Zn-SOD、T-AOC有明显降低(P<0.01);与DM组相比,Ag组能显著地降低肝脏匀浆和线粒体MDA含量(P<0.01),Ag组可提高肝脏匀浆T-AOC(P<0.05)和线粒体T-AOC(P<0.01)。与DM组相比,Ag组能降低肝脏线粒体CYP450 2E1酶活性。结论 Ag可能通过降低肝脏线粒体CYP450 2E1活性,减轻肝脏线粒体脂质过氧化反应,改善糖尿病肝脏氧化/抗氧化失衡状态,对糖尿病大鼠肝脏具有保护作用。
