摘要
目的研究龙眼壳多糖对小鼠免疫活性的影响。方法热水浸提法提取龙眼壳粗多糖,脱蛋白,脱色素,DEAE-纤维素柱层析和丙烯葡聚糖凝胶色谱S-300柱层析纯化龙眼壳多糖,硫酸-苯酚法测定多糖含量。小鼠灌胃给药15d后,MTT测定龙眼壳多糖对小鼠脾淋巴细胞的增殖能力、ELISA分析龙眼壳多糖对小鼠脾淋巴细胞分泌IL-2的作用、LDH释放法测定龙眼壳多糖对NK细胞活性的作用。结果龙眼壳多糖对正常小鼠脾淋巴细胞增殖作用实验中,剂量在250~62.5 mg/L范围内,刺激指数随着其剂量的增加而增加。龙眼壳多糖对小鼠脾淋巴细胞分泌IL-2的量,剂量在250~62.5 mg/L范围内,随着剂量的增加而增加。龙眼壳多糖对正常小鼠脾脏NK细胞活性,剂量在250~62.5 mg/L范围内,NK细胞活性随着剂量的增加而增强。结论龙眼壳多糖体内有一定的免疫作用。
Objective To study the immune activity of longan shell in vivo. Methods LP1 was extracted by hot-water and purified by deproteinizing,decolouring,DEAE-cellulose column and Sephacryl S-300 HR chromatographies.The polysaccharide content was determined by phenol-sulfuric acid method.The mice were given LP1 by gavage for 15 days,lymphocyte proliferation was detected by MTT,secretion of IL-2 was detected by ELISA,and NK cell activity was detected by LDH release. Results LP1 could promote lymphocyte poliferation,increase the secretion of IL-2 and the activity of NK cells,and the effects were dose-dependent within the dose range of 62.5~250 mg/L. Conclusion LP1 can enhance the immune system function.
