抗柯萨奇B病毒性心肌炎胶囊复方提取物对心肌保护作用的实验研究

Protective Effects of Ethacol Extract of Antitoxin Cachet for Coxsackie B Myocarditis on Myocardial Injury Induced by Adriamycin in Mice

目的观察抗柯萨奇B病毒性心肌炎胶囊复方醇提物(K-CoxBJN-C)对阿霉素(ADR)诱导小鼠心肌损伤的保护作用,并探讨其作用机制。方法选取肢体Ⅱ导联心电图正常的120只昆明小鼠随机分为6组,即正常对照组(灌胃生理盐水)、模型组(灌胃生理盐水)、水提物组(灌胃K-CoxBJN-S,5 g.kg-1)、复方65%醇提物高、中、低剂量治疗组(分别灌胃K-CoxBJN-C 1.22,.4,4.8 g.kg-1),给药1次/d,连续14 d;同时除正常对照组(隔日腹腔注射NS 3mg.kg-1)外,各组小鼠隔日腹腔注射阿霉素3 mg.kg-1,共7次,建立ADR心肌损伤模型。动态观察小鼠症状及体征、体质量变化、死亡率;末次注射ADR 24 h后取血,比色法测定血清中CK、LDH的活性;取心脏、脾、胸腺计算重量指数;取心脏观察心肌病理改变。结果 K-CoxBJN-C能显著降低小鼠血清中CK、LDH的活性(P<0.05),增加心脏、脾、胸腺重量指数,改善小鼠心功能,减少死亡率,其中醇提物高、中剂量组效果好于水煎组。结论 K-CoxBJN-C可明显减轻ADR所致小鼠的心肌损伤,其作用机制可能与减轻免疫炎性反应有关。

Objective To observe the protective effects of antitoxin cachet for Coxsackie B myocarditis on myocardium injury induced by adriamycin in mice and its mechanism. Methods 120 healthy kunming mice were randomly divided into six groups：blank group,model group,water-decoction group,HXTMLmax and HXTMLmin group,20 rats in each group.Saline was injected into mice of blank group,10 ml/kg,the rest mice were injected with adriamycin,3 mg/kg,once every 2 days,and the myocardial injury model was established after 7 times.Mice in blank and model groups were administered with saline and the rest were created with drugs by gavage,for 14 days.Symptoms and signs,weights,mortality were observed.CK and LDH in serum were determined.Weight indexes of hearts spleen and thymus were also measured：Ethanol extract pathological changes of myocardium were observed. Results The ethanol extract of antitoxin cachet for Coxsackie B myocarditis could decrease CK and LDH in serum significantly,increase the weight indexes of heart,spleen and thyrum,improve cardial function and lower mortality.HXTMLmax and middle groups had much more effects than the water-decoction group. Conclusion Alcohol extract of the drugs injury,and the possible mechanism may be related with immunal reaction decrease.
