摘要
[目的]探索应用高速逆流色谱技术从卢氏藻中分离抑菌活性化合物。[方法]采用正己烷-乙酸甲酯-乙腈-水(体积比为4∶4∶3∶4)的液-液-液3相溶剂体系为高速逆流色谱的最优分离体系,以上相和中相为固定相,依次以下相和中相对样品进行洗脱,并收集分离液经高效液相色谱检测纯度。[结果]洗脱共得到RS1、RS2、RS3 3个组分物质,经HPLC纯度检测,RS1为极性较强的单一化合物;体外抑菌试验结果表明,RS1对革兰氏阳性菌、革兰氏阴性菌及真菌都有很好的抑菌效果,RS2对细菌有抑菌效果,对真菌无抑菌作用,RS3没有抑菌活性。[结论]液-液-液3相高速逆流色谱法能够一步从卢氏藻混合物中分离获得具有良好抑菌活性的单体化合物。
[Objective] High-speed countercurrent chromatography(HSCCC) was applied to the separation and purification of the active substance from R.spectabilis.[Method] A three-phase solvent system of n-hexane,ethyl acetate,acetonitrile and water(4∶4∶3∶4,V/V) was selected,and the upper phase and middle phase was used as stationary phase.The samples were eluted by down phase and middle phase in turn,and the pure of compounds were analyzed by HPLC.[Result]Three substances(RS1,RS2,RS3) were separated,and RS1 was a monomer compound.The results of antibacterial experiments showed that RS1 had good antibacterial effect for bacteria and fungi;RS2 could only inhibit bacteria,and had no anti-fungi effect;RS3 had no antibacterial activity.[Conclusion]Three-layer HSCCC could effectively separate antibacterial monomer compounds from R.spectabilis by one step.
