PAVHB的制备及其pH敏感降解性能研究

摘要 目的合成具有pH敏感降解性的新型载体聚丙烯酰共乙烯吡咯烷酮氧肟酸酯[poly(acryl-co-vinylpyrroli-done)hydroxamic benzyl ester,PAVHB],制备口服肠溶给药系统。方法采用聚丙烯酰胺-co-N-乙烯吡咯烷酮(PAV)侧基肟化酯化的方法,合成具有pH敏感性降解性的新型材料PAVHB;通过1H-NMR、元素分析等方法表征该聚合材料的结构;紫外分光光度法测定其的降解性能。结果 PAVHB的降解具有强烈的pH敏感性,且其降解速度在生理pH附近发生突跃。结论具有pH敏感降解性的PAVHB有望成为新的口服肠溶载体材料。 Objective To establish a new enteric drug delivery system using pH-sensitive-degradation poly(acryl-co-vinylpyrrolidone) hydroxamic benzyl ester(PAVHB) carrier.Methods PAVHB was synthesized by hydroxamation and esterification of poly(acrylamide-co-vinylpyrrolidone)(PAV) side groups.1H-NMR and elemental analysis were applied to characterize this polymer.Results PAVHB exhibited a strong pH-sensitive-degradation property.The rate of degradation enhanced dramatically when the pH value was near its physiological range.Conclusion pH-sensitive PAVHB is a potential enteric drug carrier for oral administration.
