活血清解汤对高脂血症性胰腺炎大鼠瘦素的调控作用 被引量:4

摘要 目的探讨活血清解汤对高脂血症性胰腺炎(HLAP)大鼠瘦素的调控作用及其机理。方法将60只SD雄性大鼠随机分为3组:正常对照组(NC组),高脂血症性胰腺炎组(HLAP组),活血清解汤组(HXQJ组)。采用高脂饲料喂养3周后经腹腔注射L-精氨酸500 mg/100 g体重制作大鼠高脂血症性胰腺炎模型,经注胃管给予生理盐水或中药1 mL/100 g体重,每12 h一次。各组分别造模后24 h、36 h处死半数大鼠,观察血清淀粉酶、瘦素、TNF-α、IL-1β、IL-10水平的释放以及胰腺病理学、胰腺组织中瘦素和瘦素受体表达的变化。结果造模后24 h HLAP组和HXQJ组血清淀粉酶、瘦素和IL-1β、TNF-α水平均较NC组升高(P<0.01),胰腺组织损害也明显增强(P<0.01),胰腺组织瘦素表达明显增高(P<0.01)。HXQJ组淀粉酶、IL-1β、TNF-α水平均较HLAP组低(P<0.01),而瘦素和IL-10水平以及瘦素、瘦素受体表达高于HLAP组(P<0.01)。造模后36 h,HLAP组和HXQJ组淀粉酶、瘦素水平均下降,胰腺瘦素、瘦素受体表达也减弱(P<0.01),但HXQJ组淀粉酶水平已回落至NC组水平,而瘦素水平仍高于HLAP组,HLAP组和HXQJ组IL-1β、TNF-α及IL-10水平继续上升,胰腺病理损害进一步加重,但HXQJ组IL-1β、TNF-α水平较HLAP组低,而胰腺组织瘦素表达和IL-10高于HLAP组,胰腺损害也较轻(P<0.01)。结论活血清解汤能够通过上调瘦素及瘦素受体表达,调节细胞因子网络平衡,阻断炎症介质的级联效应,从而阻止HLAP的发生和发展。 Objective To evaluate the efficacy of the HuoXueQingJie decoction in adjusting the leptin releasing in rats with hyperlipidemic acute pancreatitis(HLAP).Methods Sixty SD male rats were randomly divided into 3 groups with 20 rats in each one: normal control group(NC),hyperlipidemic acute pancreatitis group(HLAP) and treatment group with HuoXueQingJie decoction(HXQJ).The HLAP model was established by single intraperitoneal injection of 500 mg/100 g body weight of L-arginine after being fed with high-fat diet for three weeks.The medicine or normal saline were given by way of gastrotube with 1 mL/100 g body weight every 12 h.Half of animals in each group were killed at 24 and 36 h after injection.Blood samples were taken for the examination of serum amylase,leptin,interleukin(IL)-1β,tumor necrosis factor(TNF)-α,IL-10 while pancreatic tissues were removed for leptin and leptin receptor expression and localization and histological evaluation.Results At 24 h after injection,the level of amylase,IL-1β,TNF-α and leptin in HLAP group and HXQJ group were higher than those in NC group(P〈0.01),but the level in HXQJ group were lower than that in HLAP group.The leptin and leptin receptor expression of pancreatic tissues in both HLAP and HXQJ groups were higher compared to those in the NC group(P〈0.01),as the expression of those in HXQJ group was higher than that of HLAP group(P〈0.01).At the same time,the damage in pancreatic tissue was significantly enhanced(P〈0.01) and the increase of IL-10 was observed in HXQJ group only(P〈0.01).After 36 hours,the level of amylase and leptin in HLAP group and HXQJ group went down.The level of amylase in HXQJ group was lower than that of HLAP group,but the level of leptin in HXQJ group was higher than that in HLAP group.In both HLAP group and HXQJ group,the level of IL-1β,TNF-α and IL-10 continued to rise and the pancreatic damage aggravated.The level of IL-1β and TNF-α in HXQJ group were lower than those in HLAP,but the level of IL-10 was higher(P〈0.01).The pancreatic damage in histology was also less P〈0.01).The leptin and leptin receptor expression decreased in HLAP group and HXQJ group but the HXQJ group was higher than HLAP group(P〈0.01).Conclusion The Chinese traditional herbs HuoXueQingJie decoction played a variety of roles in HL-AP,such as increasing the level of leptin,keeping cytokine network in balance,and blocking the cascade of inflammatory mediators.
