摘要
目的:探索一种快速、微创且能大批量制作弥漫型兔VX2肺癌模型的方法,为展开具有肺靶向抗肺癌药物及其制剂的研究奠定基础。方法:采用微创胸腔穿刺接种肿瘤的方法,将兔VX2肿瘤组织块移植于兔的右肺。采用螺旋CT技术检测VX2肿瘤在兔肺部的生长情况,并结合大体解剖及组织病理学检查评价模型的成功率以及观察肿瘤生长、转移等生物学行为。结果:该模型成功率为100%。VX2肿瘤在兔右肺的成瘤时间为5~10 d;其右胸腔内播散转移时间为20~25 d,左肺、肝及肾等转移时间为30~34 d,荷瘤兔中位生存期为32 d。结论:该方法具有简便快速、易于大批量制作、术后不良反应轻微以及模型成功率高等优点,利用本方法所制得的兔VX2肺癌模型与人类原发性肺癌有着相似的生物学行为,这可满足肺靶向抗肺癌药物及其制剂药效学研究之需。
Objective:To explore a minimally invasive surgery technique for establishing rabbit VX2 diffuse lung cancer model,so as to lay a foundation for the evaluation of the effect of lung-targeting anticancer drugs.Methods: The VX2 tumor blocks were implanted into the pulmonary parenchyma of rabbit via pleuracentesis.Spiral computer tomography,gross anatomy and histopathology examination were performed to evaluate the success rate of the model and the biological behavior of the tumor.Results: The rabbit VX2 diffuse lung cancer model was established successfully,and the success rate was 100%.The tumor formed in right lung from 5 to10 days.Right thoracic cavity was invaded by tumor from 20 to 25 days,and the metastasis was observed in left lung and liver or kidney from 30 to 34 days.The median survival time of rabbits was 32 days.Conclusion: Tumor blocks implantation via pleuracentesis is a simple and effective method to make rabbit VX2 diffuse lung cancer model with a high success rate.The adverse effect of the operation is mild.The VX2 tumor and human primary lung cancer have similar features,so the model is suitable for evaluating the effect of lung-targeting anticancer drugs.
