糖尿病肾病大鼠肾组织α-actinin-4表达及通络泄浊方的干预作用 被引量:10

摘要 目的:探讨α-actinin-4在糖尿病肾病大鼠肾组织中表达的变化,并观察通络泄浊方对其表达的影响。方法:采用链脲佐菌素(STZ)法制作大鼠模型,随机分为正常对照组、模型组、通络泄浊方组、科素亚组和通络泄浊方+科素亚组。采用Realtime-PCR及Westernblot方法检测肾组织α-actinin-4mRNA和蛋白的表达。结果:与正常对照组相比,模型组mRNA表达和蛋白明显降低(P<0.01),给药组与模型组相比,mRNA和蛋白表达明显增高(P<0.01)。结论:α-acti-nin-4的表达降低参与了糖尿病肾病大鼠蛋白尿的发生;通络泄浊方可以通过上调DN大鼠肾组织α-actinin-4mRNA和蛋白的表达而改善大鼠的肾功能及蛋白尿水平。 Objective:To investigate α-actinin-4 expression and the effects of Tongluoxiezhuofang intervention in rats with diabetes nephropathy.Methods:streptozotocin(STZ)-induced diabetic rats were established,rats were randomized into five groups:the normal group,model group,Tongluoxiezhuofang treatment group,losartan treatment group,Tongluoxiezhuofang combined losartan treatment group.Realtime-PCR and Westernblot were employed to study the change of α-actinin-4 mRNA and protein.Results:The level of expression of α-actinin-4 mRNA and protein in the model group were less than that in the normal group markedly(P〈0.01),but the level of expression in treatment group were more than the model group(P〈0.01).Conclusion:There was close relationship between the expression of α-actinin-4 with diabetic nephropathy and urine protein.It is initially confirmed that Tongluoxiezhuofang can up-regulate the expression of α-actinin-4 and reduce the urine protein on diabetic nephropathy rats.
