摘要
目的:观察不同浓度的黄芩苷对TGF-β1诱导的人近端肾小管上皮细胞(HK-2)细胞转分化的影响。方法:以5ng/ml的TGF-β1作用HK-2不同的时间(0、12、24、48和72h),用RT-PCR法检测α-平滑肌肌动蛋白、E-钙黏蛋白基因表达情况。以40、80、160、320μmol/L浓度的黄芩苷与5ng/mlTGF-β1共孵育48h,每12h在倒置显微镜下观察细胞形态。结果:黄芩苷干预后可抑制HK-2梭形变,抑制α-平滑肌肌动蛋白基因表达,上调E-钙黏蛋白基因表达,且在一定范围内呈剂量依赖性。结论:一定浓度的黄芩苷在体外有阻滞TGF-β1诱导的HK-2细胞转分化作用。
Objective:To abserve the effects of baicalin on tubular epithelial cell transdifferentiation induced by TGF-β1.Methods:The human HK-2 cells were treated by TGF-β1(5 ng/ml) atdifferent ti me points(0,12,24,48 and 72 h).The expressions of α-mooth muscle actin(α-SMA)and E-cadherin(E-cad)were detected by using RT-PCR.The HK-2 cells were be in stimulus duration 48 h in a medium contained baicalin in varying concentration(40,80,160,320 μmol/L)and 5ng/ml TGF-β1.Results:After the intervention of Baicalin,the morphology changing of HK-2 cells and expressions of α-SMA were inhibited significantly.The expressionof E-cad was upregulation.Conclusion:Baicalin treatment can abolish TGF-β1-induced EMT of HK-2 cells.
