摘要
目的研究鼠尾草酸(carnosic acid,CA)与1,25-二羟基维生素D3〔1,25(OH)2D3〕联合应用对NB4细胞生长及凋亡、分化的诱导作用。方法通过四氮唑蓝(MTT)法观察细胞的生长,光学显微镜观察细胞形态,应用流式细胞仪测定细胞周期、凋亡率及CD14的表达,观察联合应用鼠尾草酸与低浓度1,25(OH)2D3对NB4细胞的影响。结果 CA和1,25(OH)2D3都能抑制NB4细胞增殖,且联合使用抑制作用显著。CA与低浓度1,25(OH)2D3联合处理NB4细胞72 h后,细胞形态具有成熟单核细胞特征、细胞阻滞于G0/G1期、CD14的表达率为65.25%,其作用效应与对照组比较有显著性差异(P<0.01),与高浓度1,25(OH)2D3组比较作用相似或增强。结论鼠尾草酸可增强1,25(OH)2D3对NB4细胞的诱导分化及抑制细胞增殖的作用。
Objective To study the effects of carnosic acid combined with 1,25-dihydroxyvitamin D3 on the growth,apoptosis and differentiation of human acute promylocytic leukemia(APL)cell line NB4.Methods The proliferation of NB4 cells was examined by MTT assay.The changes of morphologic features of NB4 cells were observed by microscope;meanwhile,cell cycle parameters and monocytic differentiation marker CD14 were detected by flow cytometry.Results After exposed to carnosic acid or/and 1,25-dihydroxyvitamin D3,the cell proliferation was inhibited and the inhibitive effects were most significant in collaborated group.After treated with carnosic acid and lower concentrations of 1,25-dihydroxyvitamin D3 for 72 h,NB4 cells showed certain mature monocytic features.Besides,cell cycle was arrested in G0/G1 phase and the expression of CD14 was 65.25%.The beneficial effects exhibited a significant difference compared with control group,with a similar or enhanced efficacy to the upper concentrations of 1,25-dihydroxyvitamin D3.Conclusion Carnosic acid can enhance the differentiation and inhibit the proliferation of NB4 cells induced by 1,25-dihydroxyvitamin D3.
