摘要
目的研究T细胞在小鼠脊髓损伤(SCI)与修复中的作用。方法用BALB/C-NU小鼠(N组)、BALB/C-NU同胞小鼠(H组)、ICR小鼠(I组)建立不完全SCI模型,每组21只。术前、术后1、3、7、14、28、42、56d行脊髓运动功能(BBB)评分,免疫组化染色检测损伤区脊髓CD3、CD68和神经生长因子(NGF)表达。结果 H、I组在术后各时间点BBB评分均低于N组(P<0.05或P<0.01);除术后14d,H组和I组间BBB评分差异无统计学意义(P>0.05)。SCI后,各组CD3、CD68和NGF阳性细胞表达均上调;其中,H、I组CD3、NGF表达明显高于N组(P<0.01)。结论早期T细胞对SCI可能有损伤作用,而后期可能有促进修复作用。
Objective To investigate the effect of T cells on the spinal cord injury(SCI) and repair in rats.Methods BALB/C-NU mice(group N),heterozygote of BALB/C-NU mice(group H) and ICR mice(group I) with 21 rats each were used to establish the incomplete SCI models.Before operation and on the 1st,3rd,7th,14th,28th,42nd and 56th day after operation,Basso Beattie and Bresnahan(BBB) score was evaluated,and the expressions of CD3,CD68 and nerve growth factor(NGF) in the injury area of the spinal cord were detected by immunohistochemistry staining.ResultsAfter operation,BBB scores in groups of H and I were lower than those in group N(P〈0.05 or P〈0.01).Whereas except for that on the 14th day after operation,there was no significant difference in BBB score between group H and group I(P〉0.05).After SCI,the positive expressions of CD3,CD68 and NGF were all up-regulated,and the expressions of CD3 and NGF in groups of H and I were obviously higher than those in group N(P〈0.01).Conclusion T cells may damage the spinal cord in the earlier stage,but promote repair in the later stage.
