摘要
目的观察穿膜融合多肽TAT-N24对胃癌细胞MGC803迁移的影响。方法用纯化的融合多肽TAT-N24处理胃癌MGC803细胞,通过细胞迁移实验检测其对细胞运动迁移能力的影响,明胶酶谱实验观察其对肿瘤转移相关基因MMP9表达的影响。结果经TAT-N24多肽处理的MGC803细胞,其细胞迁移能力明显低于对照组细胞,差异具有统计学意义(P<0.05),但其对肿瘤转移相关基因MMP9的表达和分泌却没有影响。结论融合多肽TAT-N24能够抑制胃癌MGC803细胞的运动迁移能力,其在胃癌的治疗上可能具有潜在的应用前景。
Objective To observe the effects of cell - permeable TAT - N24 fusion peptide derived from p553, regulatory subunit of pbosphoinositide -3 kinase on cell migration of MGCS03 gastric carcinoma cells. Methods Gastric cancer cells MGC803 was treated with the purified fusion peptide TAT - N24. The ability of cell migration was detected by transwell migration assay. Expression of tumor metastasis associated gene MMP9 was observed by gelatin zymography. Results TAT - N24 fusion protein markedly resulted in decrease of the MGC803 cell migration compared with the control cells (P 〈 0.05 ). But it did not affect the expression and secretion of MMP9. Conclusion TAT - N24 fusion peptide could inhibit cell migration in gastric carcinoma MGC803 cells. It perhaps has potential clinical application in gastric carcinoma.
