CD44、NFYB及HMGB1基因表达与乳腺癌术后复发关系的研究 被引量:1

A study of the relationship of the gene expression of CD44, NFYB and HMGB1 with postoperative recurrence of breast cancer by real -time quantitative RT -PCR array

摘要 目的 研究黏附分子CD44、核转录因子Y,β(NFYB)及高迁移率蛋白B1(high-mobility group box-1,HMGB1)基因表达与乳腺癌术后复发转移的关系,初步探讨其在判断乳腺癌预后的价值.方法 81例乳腺癌患者入选,其中41例术后5年随访期内出现复发者为研究组;40例未复发仍存活者为对照组.提取所有病例石蜡包埋组织RNA,质量鉴定后,应用实时定量逆转录-聚合酶链反应(RT-PCR)芯片技术检测CD44 、NFYB及 HMGB1基因的表达,分析2组间基因表达差异,进行多因素分析.结果 研究组与对照组相比较,CD44 、NFYB及 HMGB1基因表达有差异,表达上调(P〈0.05).将基因表达与术后复发时间进行COX分析,结果显示CD44 基因表达与乳腺癌术后复发时间具有负相关(B=-0.042,Exp(B)=0.959),NFYB及HMGB1基因表达与乳腺癌术后复发时间无相关性,结果无统计学意义.结论 CD44 、NFYB及 HMGB1基因在乳腺癌术后复发组和未复发组中差异表达;基因CD44 过表达是乳腺癌预后差的指标之一,进一步研究可为预防和控制乳腺癌的复发提供新线索. Objective To investigate the relationship of the gene expressions of cell adhesion molecule CD44, nuclear transcription factor Y, [3 (NFYB) and high - mobility group box - 1 ( HMGB1 ) with breast cancer recurrence and metastasis, and to explore its significance in the prognosis of breast cancer. Methods 81 cases of breast cancer were enrolled, of which 41 cases with recurrence during 5 - year follow - up period were designated as the study group, and 40 cases still alive without recurrence as the control group. The gene expressions of CD44, NFYB and HMGB1 were determined by real - time PCR following the extraction of the RNA from paraffin embedded tissue and quality identification in all cases, and the differences in the genes expressions between the two groups were analyzed by multivariate analysis. Result Compared with the control group, the gene expressions of CD44, NFYB and HMGB1 in the study group had significant differences and were upregulated ( P 〈 0.05 ). COX analysis of the gene expressions and postoperative recurrence time revealed that the gene expression of CD44 was negatively correlated with postoperative recurrence of breast cancer ( B = -0. 042, Exp (B) = 0. 959), while the gene expressions of NFYB and HMGB1 had no correlation with the postoperative recurrence time of breast cancer, with no statistical significance. Conclusion There are significant differences in the gene expressions of CD44, NFYB and HMGB1 between breast cancer recurrence group and the non - recurrence group; the overexpression of CD44 is one of the indicators of poor prognosis of breast cancer, and further research is required so as to provide new clues to the prevention and control of breast cancer recurrence.
