摘要
目的 研究槲皮素对人宫颈癌HeLa细胞增殖及凋亡的影响,同时探讨槲皮素对抗宫颈癌的可能作用机制.方法 四唑盐比色法(MTT法)检测不同浓度及不同时间槲皮素对HeLa细胞增殖的影响;流式细胞仪分析槲皮素对HeLa细胞凋亡的影响;Western blotting法检测癌细胞caspase-3蛋白水平的变化.结果 槲皮素作用于HeLa细胞后,细胞增殖水平明显下降(P〈0.05),且在一定浓度范围内呈时间、浓度依赖性(r=0.720,P〈0.01;r=0.385,P〈0.05);不同浓度槲皮素处理HeLa细胞后能诱导其凋亡,与对照组比较差异有统计学意义(P〈0.05或P〈0.01);随着槲皮素浓度的增加,caspase-3蛋白水平明显上调(P〈0.05).结论 槲皮素可显著抑制人宫颈癌HeLa 细胞增殖并诱导其凋亡,其机制可能是通过激活caspase-3蛋白表达实现的.
Objective To investigate the effects of quercetin on the proliferation and apoptosis of human cervical cancer HeLa cells in vitro, and to explore the possible mechanism of quercetin against cervical cancer. Methods The inhibitive effects of quercetin on the proliferation of HeLa cells were detected by MTT assay at different concentrations and time points ; the apoptotic percentage was analyzed by flow cytometry (FCM) ; and the variations in the expression level of caspase - 3 protein were detected by Western blotting, respectively. Results The proliferation of human HeLa cells decreased significantly following quercetin treatment, in a time - and dose - dependent manner within certain ranges of concentrations ( r = 0. 720,P 〈 0. 01 ; r = 0. 385, P 〈 0. 05 ). Quercetin at different concentrations could induce the apoptosis of HeLa cells, and the differences were statistically significant compared with the control group ( P 〈 0. 05 or P 〈 0.01). Along with the increase of quercetin concentrations, easpase - 3 protein level was markedly up - regulated ( P 〈 0. 05 ). Conclusion Quercetin can significantly inhibit the proliferation of HeLa cells and induce the apoptosis, which might be mediated via the activation of caspase -3 protein expression.
