摘要
目的:探讨实验性高胆固醇血症大鼠肝脏胆固醇转出相关基因表达的变化,进一步阐明中药防治高胆固醇血症的作用机制。方法:将30只雄性SD雄性大鼠随机分成正常组、模型组和中药组,模型组和中药组进行8周高胆固醇饲料喂养,制作成高脂血症大鼠模型,中药组在造模型同时给予中药干预。检测各组大鼠血浆TC、TG、HDL-C、LDL-C含量以及肝脏组织中的LCAT和ABCA1基因表达水平。结果:中药可以降低大鼠血浆TC、TG和LDL-c水平,升高血浆HDL-c水平;高胆固醇血症大鼠肝脏组织LCAT和ABCA1表达水平与对照组比较明显降低,中药有明显上调LCAT和ABCA1表达水平作用。结论:中药复方对高胆固醇血症大鼠的血脂水平具有调节作用,其作用机制可能是通过调节LCAT和ABCA1等基因表达,提高肝脏细胞对胆固醇的转运与排泄功能。
Objective:To study the variety of liver cholesterol transportation related gene expression of hypercholesterolemia rat and further clarify the functional mechanism of Chinese herbal on hypercholesterolemia.Method:30 male SD rat were randomly divided into normal group(NG)、model group(MG)and herbal group(HG),building the hypercholesterolemia rat(HR) model by feeding the MG and HG with hypercholesterolemia for 8 weeks,feeding the Chinese herbal to HG at the same time.Testing content of TC,TG,HDL-C,LDL-C of blood plasma and gene expression levels of LCAT and ABCA1 of liver.Result:Chinese herb could decrease the levels of TC,TG and LDL-c of rat blood plasma,increase the HDL-c level of blood plasma.The lecithin cholesterol acyl transferase(LCAT) and ATP binding cassette transporter A1(ABCA1) gene expression level of HR liver is significant lower than NG,the Chinese herbal obviously raised the LCAT and ABCA1 gene expression.Conclusion:The Chinese herbal compound could modulate the blood lipid level of HR,the functional mechanism is by modulating LCAT and ABCA1 gene expression to improve the cholesterol transportation and excretion function of liver cells.
