胰腺癌中K-ras基因突变检测及其临床意义的初步探讨 被引量:2

摘要 目的用基因测序法检测胰腺癌患者K-ras基因突变位点,初步探讨K-ras基因对胰腺癌检测的临床意义。方法筛选20例来自肝胆外科胰腺相关疾病患者手术标本,经过病理切片证实后分成良性病变对照组和胰腺癌恶性病变组共2组,每组各10例,采用普通的PCR方法扩增人类基因组中的K-ras基因,并采用ABI 3100 DNA测序仪对扩增的片段进行测序分析,检测良性病变对照组和胰腺癌恶性病变组标本突变情况,用SPSS统计学软件计算K-ras基因突变率与胰腺癌恶性病变的相关性。结果良性病变对照组标本K-ras基因第12位、13位密码子没有发生突变;胰腺癌恶性病变组有8例发生突变(80%),其中7例胰腺标本K-ras基因的第12位密码子发生突变,1例胰腺标本K-ras基因的第13位密码子发生突变。结论在实验室建立基因测序法检测胰腺癌患者K-ras基因突变位点的方法成功,可以推广使用;胰腺癌恶性病变组织K-ras基因的突变与胰腺癌有显著相关性,且第12位密码子为突变的热点位置。
