rhTGF-β1对大鼠髁突软骨细胞SZP表达的影响

Effects of rhTGF-β1 on the SZP Expression and Secretion in the Chondrocytes Derived from Rat Condyle Cartilage

摘要 目的:观察rhTGF-β1对大鼠髁突软骨细胞SZP表达的影响。方法:酶消化法获取大鼠髁突软骨细胞,甲苯胺蓝和II型胶原染色鉴定。细胞培养液中分别加入10μg/L、20μg/L rhTGF-β1后,不同时间点应用RT-PCR和ELISA法检测SZP mRNA和蛋白的表达。结果:加入10μg/L、20μg/L rhTGF-β1 24h后髁突软骨细胞中SZP mRNA及蛋白表达均增高,且具有统计学差异(P<0.05)。加入10μg/L rhTGF-β1 24h、48h、72h后SZP mRNA及蛋白表达逐渐增强,且具有统计学差异(P<0.05)。结论:rhTGF-β1能够增强大鼠髁突软骨细胞SZP的合成和分泌,并具有时间依赖性。 Objective: To investigate the effects of transforming growth factor-β1(TGF-β1) on the expression and secretion of superficial zone protein(SZP) in the articular chondrocytes derived from rat condyle.Methods: The chondrocytes were isolated from rat mandibular condyle by enzyme digestion and phenotypically determined by toluidine blue staining and collagen II immunohistochemistry staining.After exposed to rhTGF-β1(10 and 20ng/ml) for indicated time course,the SZP mRNA expression and secretion by chondrocytes were assessed by RT-PCR and ELISA,respectively.Results: Treatment of chondrocytes with rhTGF-β1 resulted in significant enhancement in the mRNA expression and secretion of SZP as compared with control(P0.05).Moreover,rhTGF-β1(10 ng/ml) exposure remarkably induced the expression of SZP over a 72 hour time course.Conclusion: TGF-β1 can enhance the SZP expression and secretion in cultured condylar chondrocytes in a time-dependent manner.
