摘要
目的探讨骨髓干细胞(MSCs)移植对慢性马兜铃酸肾病(CAAN)肾脏损害的修复作用及可能机制。方法将30只雌性Wistar大鼠随机分为三组各10只。移植组及模型组予关木通灌胃制作CAAN模型,对照组饮用水灌胃。制模第12周末移植组经尾静脉注入大鼠MSCs 1 ml(2×107个细胞)/只;模型组及对照组经尾静脉注射等量生理盐水。干预第4周末采用免疫组化、免疫印迹法及RT-PCR法检测肾组织中细胞角蛋白(CK18)蛋白和CK18 mRNA表达。结果 CK18蛋白表达:移植组明显高于模型组,但明显低于对照组,P均<0.01。CK18 mRNA表达:RT-PCR法示模型组表达明显减少甚至消失,移植组明显高于模型组,但明显低于对照组,P均<0.01;免疫印迹法显示了同样的变化趋势。结论 MSCs移植可明显减轻CAAN肾损害;其机制可能为促进肾小管细胞CK18正常表达。
Objective To investigate the effects of mesenchymal stem cells(MSCs) transplantation on the reparation function and the possible mechanism of kidney injury in chronic aristolochic acid nephropathy(CAAN) rats.Methods Thirty female Wistar rats were randomly divided into 3 groups(10 rats/group).Transplanted group and model group were administered with Aristolochia Decoction to establish CAAN model,control group was administered with drinking water by gastrogavage.At the end of the 12th week,1 ml of MSCs was injected through caudal vein to the rats in transplanted group,while to those in model and control group equal volume saline was injected instead.At the end of the 4th week after interference,the expression change of protein and mRNA of CK18 were detected using immunohistochemical,RT-PCR and Western blotting.Results The protein expression of CK18 in transplanted group were remarkably higher than that in model group,but obviously lower than that in control group,all P0.01.The mRNA expression of CK18 in model group by RT-PCR were remarkably decreased,even disappeared,and which in transplanted group were remarkably higher than that in model group,but obviously lower than that in control group,all P0.01;and the Western blotting showed the same change.Conclusions The MSCs transplantation can obviously lighten kidney injury of CAAN,the mechanism may be throuth promoting the nomal expression of CK18 in tubule cell.
