摘要
目的筛选出对天然药物IHA-01的敏感肿瘤细胞,并对其抗瘤机制进行初步研究。方法体外培养12种人肿瘤细胞株,经3.125、6.25、12.5、25和50μg/ml 5个浓度IHA-01作用48 h后光镜下观察细胞株形态变化,采用CCK-8法计算IC50值(半数抑制浓度),确认敏感细胞株及最佳作用浓度。流式细胞仪检测IHA-01最佳作用浓度下敏感细胞株细胞凋亡情况及端粒酶活性。结果 5个浓度IHA-01均能抑制12种癌细胞增殖,确定结肠癌HCT-8细胞为敏感细胞(IC50值最低),最佳作用浓度为1.29μg/ml;鼻咽癌CNE-2细胞为不敏感细胞(IC50值最高)。1.29μg/ml的IHA-01作用后HCT-8细胞凋亡率明显高于、端粒酶活性明显低于CNE-2细胞(P均<0.01)。结论 IHA-01对HCT-8细胞有较强的抑制作用;其机制可能为诱导肿瘤细胞凋亡及抑制细胞端粒酶活性。
Objective To screen out sensitive tumor cell lines with traditional Chinese medicine IHA-01,and study its anti-tumor mechanism.Methods Twelve strains of human cancer cell lines were cultured in vitro,then treated with IHA-01 at 5 different concentrations for 48 hours.Cell morphology changes were observed by microscope,and IC50(half inhibitory concentration) values was obtained by CCK-8 assay to confirm the sensitive cell line;the apoptosis and telomerase activitieof HCT-8 exposed to the optimal concentration of IHA-01 were detected by flow cytometry.Results Five different concentrations of IHA-01 inhibited the proliferation of 12 strains human cancer cell lines,in which the sensitivity of colon cancer cells HCT-8 was the highest,the optimal concentration was 1.29 μg/ml;the CNE-2 cell was not sensitive.The apoptosis rate and telomerase activity of HCT-8 cell were respectively higher and lower than those of CNE-2 cell after treated with IHA-01 of 1.29 μg/ml(both P0.01).Conclusions The IHA-01 can inhibit the HCT-8 cell strongly;the mechanism maybe associated with inducing tumor cells apoptosis and inhibiting telomerase activity.
