摘要
目的探讨中华苦荬菜治疗糖尿病模型大鼠的物质基础。方法给予糖尿病模型大鼠不同中华苦荬菜提取物,测定大鼠血液糖化血红蛋白和果糖胺,以其为因变量,以中华苦荬菜提取物36个共有高效液相色谱峰的相对峰面积为自变量,代入SPSS统计软件进行相关分析。结果给予中华苦荬菜乙酸乙酯、正丁醇和水溶液提取物后,糖尿病模型大鼠糖化血红蛋白分别为(13.71±1.57)%,(13.44±1.18)%,(12.81±0.87)%,给予中华苦荬菜乙酸乙酯、正丁醇提取物后果糖胺分别为(0.23±0.01),(0.23±0.02)mmol.L-1,均显著高于模型组[分别为(15.04±0.99)%,(0.25±0.02)mmol.L-1];阳性对照药组(分别给予二甲双胍和参芪降糖颗粒)糖化血红蛋白分别为(10.77±1.60)%,(12.40±1.36)%,均高于模型组(P<0.05)。采用统计软件确定上述有效物质与中华苦荬菜各提取物的高效液相色谱峰的相关系数。结论初步确定与降低糖化血红蛋白和果糖胺作用明显相关的各色谱峰为中华苦荬菜作用的物质基础。
Objective To study the material foundation of ixeris chinesis(Thunb) Nakai for treating diabetic complications in rats. Methods The diabetic rats were treated with different extracts of ixeris chinesis(Thunb) Nakai.The glycolated hemoglobin and fructosamine were tested and which were referred as dependent variable,the relative area for 36 common peaks of extracts as independent variable for the correlation determination by SPSS software. Results After treatment,the glycolated hemoglobin in rats treated with n-butanol extracts,ethyl acetate extracts and water extracts were(13.71±1.57)%,(13.44±1.18)% and(12.81±0.87)%,respectively;the fructosamine were(0.23±0.01) and(0.23±0.02) mmol·L-1 by n-butanol extracts and ethyl acetate extracts,respectively,which were lower than that in the model and positive groups.The glycolated hemoglobin of positive control(metformin and shenqi sugar declining granule) were(10.77±1.60)% and(12.40±1.36)%,respectively,which was significantly fewer compared with the model group(P0.05).The related coefficience of the active substances and extracts of ixeris chinesis(Thunb) Nakai was determined by statistics software. Conclusion It's primarily demonstrated that the chromatographic peaks significantly related to the reduction of glycolated hemoglobin and fructosamine are the material foundation of ieris chinesis(Thunb) Nakai,and the molecules corresponding to the peaks may share the same or similar functional groups that are medium polar molecules.
