原儿茶酸与拉米夫定体外联合抗HBV药效学研究 被引量:10

Pharmacodynamic study of lamivudine combined with protocatechuic acid against hepatitis B virus in vitro

摘要 为观察拉米夫定(3TC)与原儿茶酸(PA)联合应用抗乙型肝炎病毒(HBV)的作用,在HepG2.2.15细胞培养液中分别加入适当浓度的3TC、PA、3TC+PA溶液,每4 d换用含相同药物浓度的新鲜培养液,第8天末取上清液,用酶联免疫吸附测定法(ELISA)检测上清液中HbsAg和HbeAg的量,用荧光定量PCR(FQ-PCR)检测培养液中HBV DNA的拷贝数,计算不同药物处理对HepG2.2.15细胞分泌HbsAg、HbeAg及HBV DNA的抑制率,并用金正均Q值法分析联合用药的抗HBV效果,确定联合用药的最佳配比及给药剂量.结果表明,与单用PA或3TC组相比,当PA∶3TC≥100时,联合用药组对HepG2.2.15细胞HbeAg和HBV DNA有相加抑制作用,对HbsAg有协同抑制作用. To evaluate the inhibitory action of lamivudine(3TC) combined with protocatechuic acid(PA) against hepatitis B virus in vitro,HepG2.2.15 cell was cultured with diffenrent concentration of 3TC,PA,3TC+PA,respectively.The culture fluid of HepG2.2.15 cell was replaced with the fresh culture medium containing the same content of the drug every 4 d.At the 8th day,HbsAg,HbeAg and HBV DNA in the culture media were detected.Jin Zhengjun Method was applied to evaluate the drug-drug interaction of 3TC and PA against HBV.The results indicated that the combination group of 3TC and PA had an additive inhibitory action on HbeAg and HBV DNA,a synergistic inhibitory action on HbsAg,respectively when the mass concentration ratio of PA versus 3TC was ≥100.
