HPLC-ELSD法测定参芪升阳补血胶囊中黄芪甲苷的含量

Determination of the content of astragaloside Ⅳ in Shenqishengyangbuxue Capsules by HPLC-ELSD

目的建立参芪升阳补血胶囊中黄芪甲苷的含量测定方法。方法以黄芪甲苷为测定指标,用HPLC-ELSD法测定,色谱条件为:色谱柱为Kromasil C18(250mm×4.6mm,5μm),流动相为乙腈-水(38∶62),流速为0.8mL.min-1;ELSD漂移管温度设置100℃,压缩空气流速2.5mL.min-1。结果黄芪甲苷对照品线性范围为0.458～13.74μg,回归方程为Y=1.376 1 X+5.961 7,r=0.999 9,平均回收率为98.2%,RSD为3.1%。结论该方法简便、灵敏,结果准确,可作为参芪升阳补血胶囊中黄芪甲苷的含量测定方法。

Objective To establish a determination method for astragaloside Ⅳ in Shenqishengyangbuxue Capsules by HPLC-ELSD.Methods HPLC-ELSD was used in the quantitative analysis by using Kromasil C18 chromatography column（250 mm×4.6 mm,5 μm） and acetonitrile-water.（38∶62） as a mobile phase.The flow rate of mobile phase was 0.8 mL·min-1.The tube temperature of the detector was 100 ℃.The flow rate of air was 2.5 mL·min-1.Results The standard curves of astragaloside Ⅳ showed a good linearity from 0.458 to 13.74 μg,and the regression equation was Y=1.376 1X＋5.961 7,r=0.999 9.The average recovery was 98.2%,with 3.1%.Conclusion The method is simple,sensitive and accurate,and can be used as the quantity control method of Shenqishengyangbuxue Capsules.
