摘要
目的研究白假丝酵母菌感染小鼠脾细胞IL-2mRNA的表达,从分子水平探讨感染小鼠的细胞免疫机制。方法用107 CFU/ml白假丝酵母菌经尾静脉注射小鼠0.2ml/只,健康对照组注射同体积无菌生理盐水。两周后,采用RT-PCR方法半定量检测小鼠脾细胞IL-2mRNA水平。结果 RT-PCR检测小鼠脾细胞均表达IL-2mRNA,扩增片段大小为477bp;感染组小鼠脾细胞IL-2与β-actin光强度比值为0.39±0.12,健康对照组为0.22±0.11,差异有统计学意义(t=2.502,P<0.05)。结论白假丝酵母菌感染小鼠后,可引起小鼠机体IL-2mRNA表达水平上调,并可能进一步引起机体细胞免疫功能的改变,从而对真菌的感染做出应答。
Objective To investigate the transcription of IL-2 in spleen cells of mice infected with Saccharomyces albicans and to explore the molecular mechanisms of cell-mediated immunity in the infected mice.Methods Mice in an experimental group were intravenously injected with 0.2 ml of 107CFU/ml S.albicans in the tail while healthy mice in the control group were administered the same volume of sterile saline.IL-2 mRNA in spleen cells was semiquantitatively analyzed by reverse transcription polymerase chain reaction(RT-PCR) two weeks after infection.Results The expression of IL-2 mRNA in mice spleen cells was detected by RT-PCR,and the amplified fragment had 477 bp.The ratio of IL-2/β-actin in spleen cells of mice in the infected group was 0.39±0.12 and that in spleen cells of mice in the control group was 0.22±0.11(t=2.502,P〈0.05).Conclusion The level of expression of IL-2 mRNA in mice infected with S.albicans rose.Infection with S.albicans may have changed the cellular immune function in infected mice and then induced an immune response to the fungal infection.
