人巨细胞病毒感染及Bcl-2表达与结直肠癌相关性研究 被引量:2

Correlation between human cytomegalovirus infection and expression of the Bcl-2 gene in colorectal cancer

摘要 目的探讨人巨细胞病毒(human cytomegalovirus,HCMV)感染在结直肠癌发生、发展过程中可能的作用机制。方法收集50例结直肠癌患者的病理肿瘤组织标本,同时收集距离肿瘤组织手术切缘10cm以上的自身瘤旁正常肠粘膜组织为对照,应用RT-PCR技术检测组织中HCMV的感染情况,免疫组化技术检测凋亡相关基因Bcl-2的表达水平。结果肿瘤组及瘤旁组HCMV IE2阳性率分别为40.00%(20/50)和6.00%(3/50),差异有统计学意义(P<0.05);Bcl-2阳性率分别为36.00%(18/50)和8.00%(4/50),差异有统计学意义(P<0.05)。50例HCMV阳性肿瘤组织Bcl-2阳性率高于HCMV阴性肿瘤组织,肿瘤组织Bcl-2表达水平与HCMV感染呈线性相关(χ2=5.834,P<0.05)。结论 HCMV感染是结直肠癌发生、发展的相关因素之一,其作用机制与影响Bcl-2的表达有关。 Objective To investigate the role of HCMV infection in the development of colorectal cancer and its possible mechanisms.Methods Tumor tissue samples were collected from 50 patients with colorectal cancer and samples of normal tissue 10 cm distant from surgical margins were collected to serve as controls.HCMV infection was detected by RT-PCR in tumor tissue and tumor-adjacent normal tissue,while the expression of Bcl-2 was evaluated with immunohistochemical straining.Results The rate of positivity for HCMV IE2 was 40%(20/50) in tumor tissue and 6%(3/50) in adjacent normal tissue;the difference in the two was statistically significant(P〈0.05).The rate of positivity for Bcl-2 was 36%(18/50) in tumor tissue and 8%(4/50) in adjacent normal tissue;the difference between the two was statistically significant(P〈0.05).Positivity for Bcl-2 was higher in the 50 samples of HCMV-positive tumor tissue than in the HCMV-negative tumor tissue,and the level of Bcl-2 in tumor tissue correlated with HCMV infection(χ^2=5.834,P〈0.05).Conclusion HCMV infection is related to the development of colorectal cancer and a possible mechanism may be its effect on the expression of Bcl-2.
