摘要
目的探讨电化学疗法对人乳腺癌MCF-7细胞凋亡及周期的影响。方法将等量人肿瘤细胞接种于培养板中,随机分为对照组和治疗组,治疗组按电量不同又分4组,对照组不通电。电化学治疗后培养6h和24h后用流式细胞仪分别检测各组细胞周期变化及凋亡率。结果电化学治疗后6h和24h MCF-7细胞凋亡率治疗组比对照组增加明显(P<0.05);治疗各电量组随电量的增加处于G0/G1期的细胞比例先逐渐增高,但至20C时反降低;而S期细胞比例有随电量增加下降至20C时升高的趋势。结论电化学疗法可抑制体外培养的人乳腺癌MCF-7细胞株的生长,其机制与诱导肿瘤细胞凋亡及影响其细胞周期有关。
Objective To explore the effect of electrochemical therapy on apoptosis and cell cycle of human breast cancer cell line MCF-7.Methods Equal number of breast cancer cells were put into Cell Culture Cluster.They were divided into control group and treated group randomly.Treated group was subdivided into 4 groups according to the different charge.6 hours and 24 hours after treatment,the rate of apoptosis and the percentage of cell cycle were detected by flow cytometry.Results 6 hours and 24 hours after treatment: the rate of MCF-7 apoptosis was higher in treated group than that in control group(P0.05);treatment with the power of the increase in electricity group G0/G1 phase ratio of cells was higher,but first to 20 C against lower when;and S phase ratio increased with the power cells down to 20 C rise.Conclusion Electrochemical through apoptosis and cell cycle can inhibit the proliferation of human breast cancer cell line MCF-7growth.
