大鼠急性血瘀模型建立及复方丹参滴丸化瘀作用研究 被引量:6

摘要 目的建立最佳剂量肾上腺素(Adrenaline,AD)大鼠急性血瘀模型,并探讨复方丹参滴丸(compounddanshen dripping pills,CDDP)化瘀作用。方法大鼠背部皮下注射0.8、1.2、1.6mg.kg-1AD各2次(间隔4h)。在2次间,将大鼠置冰水中5min,观测血瘀指标;观测不同剂量CDDP(160、80、40mg.kg-1)对血瘀指标的影响。结果注射0.8mg.kg-1和1.2mg.kg-1AD可显著增加大鼠全血黏度、血浆黏度、卡森黏度值(P<0.01);1.2mg.kg-1AD还可以显著增加红细胞电泳时间(P<0.01)和红细胞压积(P<0.05);CDDP 80mg.kg-1可显著改善模型大鼠血瘀指标(P<0.05)。结论间隔4h注射AD1.2mg.kg-1,两次间加冰水浴5min为最佳血瘀造模方案;CDDP80mg.kg-1具有改善急性血瘀的作用。 Objective To establish the model of acute blood stasis by the best dosage of adrenaline(AD),and to explore the preventive effect of Compound Danshen Dripping Pills(CDDP)on blood stasis.Methods The rats were injected with AD subcutaneously 0.8,1.2,1.6mg·kg-1,4h and then added the same dose of epinephrine.Between the 2 times(before and after the interval 2h),the rats were placed in ice water for 5 minutes,the effects of different doses of AD on the impact of hemorheology;after determine the optimal dose of epinephrine,comparing the effects of different doses of CDDP(160mg·kg-1,80 mg·kg-1,40mg·kg-1) on the hemorheology of rats.Results Injection of 0.8mg·kg-1 and 1.2mg·kg-1 AD significantly increased whole blood viscosity,plasma viscosity and Carson viscosity(P0.01).1.2mg·kg-1 AD could increase erythrocyte electrophoresis time(P0.01)and hematocrit(P0.05) significantly;80 mg·kg-1 CDDP IG to rats significantly reduced blood viscosity,plasma viscosity,erythrocyte electrophoresis time and Casson viscosity(P0.05).Conclusion 4h interval twice 1.2mg·kg-1 epinephrine injection,intermediate ice water bath for 5 minutes is the best modeling conditions;CDDP 80 mg·kg-1 played a better role in this model.
