摘要
目的分析人水通道蛋白1的B细胞优势表位,为后续研究获得其单克隆抗体做好铺垫。方法分别对人水通道蛋白1进行亲水性、二级结构、柔韧性、极性、可及性、电荷分布等多参数进行分析,然后用抗原性指数的方法综合分析。结果人水通道蛋白1的肽段21~39、39~45、42~49、79~93、110~125、122~128、130~138、153~164、229~237预测结果具有较高的免疫原性。结论多参数预测分析获得多个人水通道蛋白1表位多肽,为后续制备抗人水通道蛋白1单克隆抗体奠定了基础。
Objective To identify the B cell dominant epitope of human aquaporin protein l ( hAQP - 1 ), and to establish basis for obtaining monoclonal antibody to hAQP - 1 in the subsequent research. Methods Multiparameters such as hydrophilicity, secondary structure,flexibility,polarity,accessibility,and charge distribution of the hAQP - 1 were synthetically analyzed by means of antigenic index method. Results The peptide of 21 - 39,39 - 45,42 - 49,79 - 93,110 - 125,122 - 128,130 - 138,153 - 164 and 229 - 237 in hAQP - 1 showed high immunogenicity. Conclusion The peptides with high immunogenicity in bAQP - 1 can be obtained through preliminary analysis of muhiparameters, which establishes the basis for subsequent preparation of monoclonal antibody to hAQP- 1.
