丙酮酸(盐)的液体发酵及其产物提取工艺优化的研究

摘要 采用TTC-CaCO3复合平板法筛选丙酮酸(盐)高产菌株,并采用正交实验优化丙酮酸(盐)的液体发酵条件及其产物提取工艺,结果表明:丙酮酸(盐)液体发酵培养的最优维生素组合为盐酸硫胺素0.015 mg/L,烟酸9 mg/L,生物素0.020 mg/L,盐酸吡哆醇0.35 mg/L;最优提取条件组合为:萃取剂种类为磷酸三正丁酯,发酵液与萃取液体积比为1:2,提取温度为20℃,其中萃取剂种类、发酵液与萃取液体积比对提取率的影响显著(P<0.05)丙酮酸(盐)发酵产物浓度及提取率均基本达到国内研究水平。 In this paper,high pyruvic acid(pyruvate) producing strain was screened by TTC-CaCO3 complexes medium.The orthogonal experiment was used to optimize the technology for liquid fermentation and product extraction of pyruvic acid(pyruvate).The results indicated that the optimal concentration of the vitamin combination for liquid fermentation of pyruvic acid(pyruvate) was determined: thiamine hydrochloride 0.015 mg/L,nicotinic acid 9 mg/L,biotin 0.020mg/L,pyridoxine hydrochloride 0.35 mg / L.The optimal condition of product extraction was as follows: tributyl phosphate was used as extractant,the ratio of fermentation broth and extractant was 1:2,and the extracting temperature was 25℃.The species of extractant and the ratio of fermentation broth and extractant had a notable impact on the product extraction(P0.05).The fermentation product concentration and extraction yield of pyruvic acid(pyruvate) matched the advanced level in our country.
