摘要
目的设计一种耐药基因检测芯片,可并行检测临床常见的革兰阴性杆菌中较常见耐药基因。方法收集临床分离的70株革兰阴性杆菌:肺炎克雷伯菌、大肠埃希菌、阴沟肠杆菌、铜绿假单胞菌、鲍氏不动杆菌,根据已知耐药基因序列设计并合成引物及探针,制备耐药基因检测芯片,进行杂交及扫描。结果耐药基因检测芯片除未检到IMP耐药基因外,其他耐药基因检测结果均与PCR检测结果一致。结论该研究设计的革兰阴性杆菌耐药基因检测芯片有较好的应用价值。
OBJECTIVE To design a resistance gene detection chip to detect common clinical drug resistance genes of Gram-negative bacteria. METHODS We collected 74 clinical isolates of Gram-negative bacilli (Klebsiella pneumoniae , Escherichia coli , Enterobacter cloacae, Pseudomonas aeruginosa , Acinetobacter baurnannii ). The primers and probes were designed according to the known sequences of drug resistance genes, resistance gene detection chips were prepared, then hybridized and scanned. RESULTS In addition to the IMP resistance gene was not detected, the other resistance gene detection results of the resistance gene detection chip are consistent with the PCR test results. CONCLUSION The design of Gram-negative bacteria resistant gene detection chip has better value.
