慢性脑缺血大鼠OXR的表达变化及天智颗粒干预的实验研究 被引量:4

摘要 目的研究慢性脑缺血病程中食欲素受体(Orexin receptor,OXR)的表达水平及天智颗粒的干预对其表达水平的影响。方法通过结扎双侧颈总动脉建立脑缺血大鼠模型,水迷宫法进行行为学评价,免疫组化法测定OXR的表达水平。结果缺血15 d时大鼠的学习记忆能力明显减退,1月、2月模型组较15d模型组好转。缺血15 d时OXR表达水平明显增高,1月时OXR的表达水平较15 d时明显下降,2月时OXR的表达水平再次增高。治疗组与其模型组相比,学习记忆能力均明显好转;治疗15 d OXR的表达水平较模型组明显下降,治疗2月OXR的表达水平较模型组明显升高。结论 Orexin系统可通过某种途径对慢性脑缺血起双向调控作用,而天智颗粒可通过orexin系统治疗脑缺血。 Objective To study the OXR expression during the course of chronic cerebral ischemia and the effects of Tianzhikeli. Methods We established the cerebral ischemic model by ligating bilateral carotid arteries and evaluated the etholgy by water maze and measured the OXR expression by immunohistochemical technique. Results The intelligence of 15 d model group decreased obviously,1month and 2-month model group improved compared with 15 d model group.At the same time,the OXR expression of 15 d model group increased significantly,1month model group decreased,however 2-month model group increased the second time.The intelligence of treatment group improved obviously compared with its corresponding model group.The OXR expression of 15d threatment group decreased obviously compared with its model group,but the expression of 2-month treatment group increased significantly. Conclusions Orexin system has two-way regulatory founction through some pathway.Tianzhikeli can treat cerebral ischemia through orexin system.
