miRNA-183下调Ezrin蛋白表达抑制成骨肉瘤细胞的侵袭和转移

Down-regulation effect of miRNA-183 on ezrin protein expression inhibits the invasion and metastasis of human osteosarcoma cells

摘要 目的探讨miRNA-183下调Ezrin蛋白表达对人成骨肉瘤细胞系MG-63细胞侵袭转移能力的影响。方法以qRT-PCR和Western blot技术检测人成骨肉瘤细胞系MG-63细胞在转染miRNA-183模拟物前后miRNA-183和Ezrin蛋白的相对表达量。同时以划痕实验和transwell实验分析转染miRNA-183模拟物对MG-63细胞迁移、侵袭能力的影响。结果与正常人胚成骨细胞hFOB细胞相比,人成骨肉瘤细胞MG-63存在miRNA-183的低表达和Ezrin蛋白的表达上调(P<0.05)。在稳定转染miRNA-183模拟物后,MG-63细胞miRNA-183丰度显著上调,并明显抑制Ezrin蛋白的表达(P<0.05)。而转染后的MG-63细胞与转染前相比,其迁移和侵袭能力也显著降低(P<0.05)。结论 MiRNA-183可以通过下调靶蛋白Ezrin表达抑制人成骨肉瘤细胞系MG-63细胞的侵袭转移能力。 Objective To study the down-regulation of miRNA-183 on ezrin protein expression and its effect on inhibition of the invasion and metastasis of human osteosarcoma cells.Methods The expressions of miRNA-183 and Ezrin protein in human osteosarcoma cell line MG-63 were quantitated by qRT-PCR and Western blot before and after tansfected miRNA-183 mimics.The migaraion and invasion of osteosarcoma cells were examined in vivo by wound healing assay and transwell assay.Results Compared to normal human fetal osteoblastic cell line hFOB cells,the expression of miRNA-183 was relatively lower,but that of Ezrin protein was higher in MG-63 cell line(P〈0.05).However,down-regulation of Ezrin expression by miRNA-183 was found after transfected miRNA-183 mimics in MG-63 cells,and the cells migaraion and invasion in vivo were significantlly inhibited(P〈0.05).Conclusion MiRNA-183 may inhibit the invasion and metastasis of tumor cells in human osteosarcoma cells by down-regulation of Ezrin protein.
