贵州省汉族、苗族及布依族人群载脂蛋白M基因多态性与血脂的相关性 被引量:2

The Relationship between Apolipoprotein M Gene Polymorphism and Serum Lipids in Han,Miao and Buyi Nationalities of Guizhou Province

摘要 目的:探讨载脂蛋白M(apo M)基因启动子区多态性在贵州省汉族、苗族及布依族人群之间的差异,并分析单核苷酸多态性对apo M水平及血脂水平的影响。方法:奥林巴斯AU5400全自动生化分析仪测定血清甘油三酯(TG)、胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)及低密度脂蛋白胆固醇(LDL-C),应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)对150例汉族、140例苗族及140例布依族人群血标本apo M启动子区域T-778C进行基因多态性分析,应用酶联免疫吸附试验(ELISA)试剂盒测定血清apo M。结果:汉族人群apoM-778位点C等位基因频率明显高于苗族人群(P=0.005)与布依族人群(P=0.005),苗族与布依族人群apoM-778位点C等位基因频率无差异,apo M-778位点CT+CC型血清apo M及HDL-C含量均低于TT型(P=0.026,P=0.048)。结论:apo M基因启动子区T-778C存在民族差异,汉族人群apo M-778位点C等位基因遗传携带率高于苗族与布依族,同时apo M-778位点C等位基因引起血清apo M水平和HDL-C水平降低。 To investigate whether there are differences in the promoter region of apolipoprotein M (apo M) gene among different nationalities, and to explore the effect of mononucleotide polymorphism on apo M levels and serum lipid levels. Methods: Triglyceride (TG), cholesterol ( TC), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) were determined with Olympus AU5400 automatic biochemistry analyzer. Polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) was employed to analyze polymorphism of apo M gene promoter region T-778C in blood preparations of 150 Han, 140 Miao, and 140 Buyi nationality healthy subjects in Guizhou province. Enzyme linked immunosorbent assay was used to measure the serum apo M levels. Results: The frequency of C allele of apo M -778 site in Han nationality was higher than those in Miao (P = 0.005 ) and Buyi (P = 0.005 ) nationalities. No difference was found in the frequencies between Miao and Buyi nationality. Serum levels of apo M and HDL-C of which apo M-778 site was CT + CC genotype were lower than those of TY genotype ( P = 0. 026, P = 0.048 ). Conclu- sions: The ethnic differences exist in apo M gene promoter region T-778C. The frequency of C allele of apo M-778 site in Han nationality is higher than that in Miao and Buyi nationalities. C allele at apo M- 778 site causes the decrease of serum apo M and HDL-C levels.
