摘要
目的预测瞬时受体电位阳离子通道蛋白6(TRPC6)的抗原表位特性,筛选合适的TRPC6氨基酸序列合成多肽,用于制备抗TRPC6合成肽单克隆抗体。方法根据TRPC6氨基酸序列,采用IEDB分析及蛋白质软件(Lasergene710WinInstall)分析,预测TRPC6表面性、抗原性、亲水性及可能的二级结构性质,以此筛选合成多肽序列。结果确定合成多肽为TRPC6的胞外段565-579氨基酸序列,其表露性预测值1.000;柔韧性预测值1.008;β-转角预测值1.022;抗原性预测1.034;亲水性预测2.043。结论所确定的TRPC6多肽序列表位预测符合要求,为TRPC6合成肽制备抗单克隆抗体提供了一条有效的途径。
Objective Predicting the antigen characteristics of short transient receptor potential channel of protein6(TRPC6) and screening appropriate synthetic polypeptide of TRPC6,in order to prepare the specific anti-TRPC6 monoclonal antibody.Methods Analysis of TRPC6 amino acid sequence by IEDB and Lasergene710WinInstall software,and prediction of epitope,antigenicity,hydrophilia and secondary structure of TRPC6.Results The synthetic polypeptide sequence was determined,which is consisted of 565-579 amino acid of TRPC6 extracellular domain.The surface accessibility,flexibility,beta-turn,antigenicity and hydrophilicity were 1.000,1.008,1.022,1.034 and 2.043,respectively.Conclusion The prediction of TRPC6 synthetic polypeptide meets the requirement for preparing anti-TRPC6 monoclonal antibody.
