摘要
目的 观察白藜芦醇对人急性淋巴细胞白血病Molt-4细胞的生长抑制及凋亡诱导作用.方法 白藜芦醇处理体外培养的Molt-4细胞后,噻唑蓝(MTT)比色法测定细胞生长抑制率,Wrigh-Gimesa染色、透射电镜观察细胞的形态变化,流式细胞术检测细胞的凋亡率及周期分布.结果 MTT结果显示,200 μmol/L的白藜芦醇作用于Molt-4细胞72 h后,细胞的生长抑制率达到74.98%,并呈时间、剂量依赖性;Wrigh-Gimesa染色和透射电镜结果显示:与对照组比较,经白藜芦醇处理后Molt-4细胞出现体积缩小,染色质浓缩边集化等典型的凋亡细胞形态学改变;流式细胞术检测结果显示100 μmol/L白藜芦醇作用于Molt-4细胞48 h后78.8%的细胞被阻滞于S期.结论 白藜芦醇能明显抑制Molt-4细胞增殖,引起细胞周期的S期阻滞,并进一步诱导其凋亡.
Objective To explore the inductive effect of resveratrol on the growth and apoptosis of human acute T lymphoblast leukemia Molt-4 cells. Methods After treatment with resveratrol,MTT assay was used to determine the growth inhibitory rate of MOLT-4 cells. After the cells was stained with Wright-Gimesa,transmission electron,microscope tech- nique was used to detect the changes of morphology. Flow cytometry was used to detect the cell cycle arrest and the apop- tosis of Moh-4 cells before and after resveratrol treatment. Results After treatment with 200 μmol/L resveratrol for 72 hours ,resveratrol was able to inhibit the proliferation of Molt-4 cells in a time-dependent and dose-dependent manner,and the inhibit rate arrived 74. 98%. Wright-Gimesa staining and transmission electron microscope manner showed that :Compared with control groups,resveratrol induced Molt-4 cells to undergoapoptosis with typical apoptotic characteristics,in- cluding morphological changes of nuclear chromation condensation and the bulk of nuclear lessening. Flow cytometry assay showed that 78. 8% of the cells were arrested at the S-phase after exposed to 100 μmo/L resveratrol. Conclusion Resver- atrol could inhibit the proliferation of Molt-4 cells evidently,cause S-phase arrest and induce the cells to apoptosis.
