摘要
采用热水浸提树舌发酵浸膏中的水溶性多糖,苯酚—硫酸法测定多糖的含量,MTT法测定GAFP对SW1116细胞的生长抑制率,荧光染色观察细胞的核形变化,流式细胞仪检测细胞周期等方法,研究了树舌发酵浸膏粗多糖(GAFP)体外对人结肠腺癌细胞SW1116的生长抑制作用。结果表明:与正常Vero细胞相比较,GAFP对SW1116细胞的生长抑制具有时间和剂量依赖性;荧光显微镜下观察到经GAFP处理的细胞呈现出凋亡早、晚期的染色质变化;流式细胞仪检测结果显示GAFP能将SW1116细胞阻滞于细胞周期的G2/M期。结论:GAFP通过影响DNA的合成和阻滞细胞周期达到体外抑制SW1116细胞生长的作用。
To explore inhibitory effect of GAFP on growth of human adenocarcinoma of colon (SWIll6) cells in vitro, GAFP was obtained through water extraction and the concentration of polysaccharides was determined with Phenol-Sulfuric acid method; using MTY to assay the inhibition ratio on growth of SW1116 ceils, the changes of nucleus were measured with fluorescence staining and flow cytometry was used to investigate cell cycle. Compared to Vero normal cells, GAFP could obviously inhibit the proliferation of SW1116 with time and dose-dependent relation; besides, chromatin changes of early and late apoptosis in nucleus with fluorescence microscopy could be observed; FACS show that GAFP could induce G2/M phase arrest in cell cycle. GAFP could inhibit the growth of SWlll6 by the way of affecting DNA synthesis, arresting mitotic cycle on G2/M phase.
