响应面法优化南瓜籽芽蛋白粉的碱法制备工艺 被引量:2

摘要 为有效利用蛋白质资源,研究将经过发芽处理的南瓜籽利用超临界CO2脱脂后,采用碱溶酸沉法提取蛋白质。采用响应面分析法,研究提取温度、液料比、提取时间、提取液pH对蛋白纯度的影响,以优化碱溶酸沉法制备南瓜籽芽蛋白粉的工艺条件。实验结果表明:制备南瓜籽芽分离蛋白的最佳条件为液料比40∶1、提取温度43℃、提取时间2 h、pH 10.6。该条件下预测蛋白纯度达到96.64%,实际测得蛋白纯度为94.79%。 For effective use of protein resources,this study used the alkali extraction followed by acid precipitation to extract the Pumpkin Seeds Sprout protein which was degreased by the supercritical carbon dioxide fluid extraction(SFE-CO2).The effect of extraction temperature,liquid/material ratio,extraction time,and alkali extraction pH on protein purity is studied,thus to optimize the technological condition of the alkali-solution extraction procedure by response surface methodology(RMS) on the basis of single factor experiments.The results indicated that the optimum conditions of extraction are: liquid/material ratio of 40∶1;extraction temperature of 43 ℃;extraction time of 2 h;and alkali extraction pH of 10.6.The purity predictive value and the true value of pumpkin seeds sprout protein isolate were 96.64% and 94.79%.
