热化疗对荷瘤鼠及口腔癌患者CTL与HSP70表达水平影响的观察 被引量:3

Observation of impact of thermo-chemotherapy on the expression levels of cytotoxic T-lymphocyte and HSP70 in tumor-bearing mice and patients with oral cancer

摘要 目的:探讨热化疗对荷瘤鼠及口腔癌患者细胞毒性T淋巴细胞(CTL)及热休克蛋白70(HSP70)表达水平的影响,并初步探讨两者之间的相关性。方法:对Balb/c鼠进行热化疗处理,检测CTL活性及HSP70表达变化情况。20例口腔癌患者接受热化疗,于治疗前后行CTL活性检测,并行免疫组化检测HSP70表达情况。结果:热化疗组CTL活性与正常鼠差异无统计学意义(t=1.78,P=0.66),而与高温治疗组、平阳霉素治疗组和不治疗组比较差异有统计学意义,P=0.00。热化疗组HSP70表达明显增多,且FCM提示差异有统计学意义,P=0.00。临床试验中热化疗后口腔癌患者CTL及HSP70较热化疗前明显升高。结论:热化疗能显著提高荷瘤宿主CTL的活性,以及提高HSP70的表达,HSP70可诱导肿瘤细胞表达CTL。 OBJECTIVE: To investigate the impact of thermo-chemotherapy on the expression levels of cytotoxic T-lymphocyte(CTL) and heat shock protein(HSP70) in tumor-bearing mice and patients with oral cancer, and investigate the correlation between CTL and HSP70. METHODS: After treatments, the expression of CTL and HSP70 were detected, 20 cases of oral cancer patients received thermo-chemotherapy, the activity of CTL was detected before and after treatment, and the expression of HSP70 was detected on immunohistochemistry. RESULTS: CTL activity of thermo-chemotherapy group were no significant different from those of normal mice (t= 1.78,P=0. 66), but they were significantly different from HS, CT and NT groups(P= 0. 00). The expression of HSP70 on thermo-chemotherapy group were significantly increased, and the flow cytometry(FCM) suggested a significant difference from others (P=0. 00). In clinical trials, the expression levels of CTL and HSP70 on oral cancer patients after thermo-ehemotherapy were significantly higher than those before thermo-chemotherapy. CONCLUSIONS: Thermochemotherapy can significantly improve the CTL activity of tumor-bearing hosts, and enhance the expression of heat shock protein 70. HSP70 can induce the expression of CTL in tumor cells.
