摘要
为了建立高效的平邑甜茶离体再生体系,以平邑甜茶子叶和下胚轴为外植体,研究了不同外植体部位、暗培养时间、基本培养基、激素种类和浓度对不定芽再生的影响。结果表明,近轴端子叶的再生频率是远轴端子叶的3.1倍、下胚轴的3.7倍。在附加不同浓度的NAA、IBA与TDZ的SC培养基上,近轴端子叶的不定芽再生率达90%以上。在附加1.0mg·L-1TDZ+0.2mg·L-1NAA的再生培养基上,平均每个外植体再生芽数达到12个。研究表明,平邑甜茶近轴端子叶具有很强的离体再生不定芽能力,培养基中附加TDZ可有效提高平邑甜茶的不定芽再生效率。
To establish a highly efficient in vitro regeneration system of Malus hupehensis var. pingyiensis, the effects of types of explants, time of culture in darkness, basic medium, kind and concentration of plant growth regulator on adventitious bud regeneration from cotyledon and hypocotyl were studied. The regeneration rate of paraxial cotyledons was 3.1 times of that of abaxial cotyledons and 3.7 times of that of hypocotyls. On the SC medium supplemented with different combinations of NAA, IBA and TDZ, the regeneration rates of paraxial cotyledons of all the treatments were more than 90%. The number of regenerated buds per explant reached 12 when the medium was supplemented with TDZ 1.0 mg.L-1 and 0.2 mg.L1 NAA. The study showed that the paraxial cotyledons of M. hupehen- sis var. pingyiensis had higher ability to regenerate adventitious bud, and TDZ supplemented in re- generation medium could increase the regeneration rate of adventitious buds.
