益气血补肝肾方对Meis1在围着床期小鼠子宫内膜中表达的影响 被引量:2

摘要 【目的】观察益气血补肝肾方对Meis1在围着床期小鼠子宫内膜中表达的影响。【方法】采用免疫组织化学法对Meis1蛋白进行定位及半定量检测;采用免疫印迹法在蛋白水平定量检测Meis1在围着床期小鼠子宫内膜中的表达。【结果】Meis1在围着床期小鼠子宫内膜中呈周期性的表达。免疫组织化学结果显示:妊娠第1天(D1)开始Meis1表达逐渐增强,D4、D5表达显著增强,着床后妊娠D6在蜕膜中的表达增强(均P<0.05)。中药组小鼠子宫内膜中Meis1在D4和D5表达增强,与正常对照组比较差异有显著性意义(P<0.05)。免疫印迹检测结果与免疫组织化学结果相似,中药组小鼠子宫内膜Meis1表达增强,但与正常对照组比较差异无显著性意义(P>0.05)。【结论】益气血补肝肾方改善子宫内膜容受性,促进胚胎着床的作用与其能增强MeisI在子宫内膜的表达有关。 Objective To observe the effect of Yiqixue Buganshen Recipe'(YBR) on Meisl expression in the endometrium of mice during the peri-implantation period.Methods Immunohistochemical method was used for Meisl protein alignment and semi-quantitation,and Western blotting method was used to detect Meisl expression level in the endometrium of mice during peri-implantation period.Results Cyclic expression of Meisl was presented in the endometrium of mice during peri-implantation period.The results of immunohistochemistry showed that Meisl expressed from the first day of gestation,the expression increased at the 4th and 5th day of the gestation,and became obvious in the decidual membranes at the 6th day of the gestation after embryo implantation(P0.05 compared with that at the first day of gestation).In YBR group,Meisl expression increased at the 4th and 5th day of the gestation P0.05 compared with that in the normal control group).The results of Western blotting showed the similar results to immunohistochemistry.Meisl expression increased in the endometrium of mice in YBR group,but the difference was insignificant as compared with that in the normal control group(P0.05).Conclusion Yiqixue Buganshen Recipe is effective to improve endometrial receptivity and plays an important role in embryo implantation,and its mechanism is probably related with the increase of Meisl expression in the endometrium of mice.
