摘要
目的:研究家蝇幼虫乙醇提取物(MDE)对酵母致炎发热大鼠的解热作用及机制。方法:利用干酵母致炎大鼠发热动物模型,设立生理盐水组:ig等体积生理盐水;模型组:ig等体积生理盐水+大鼠背部sc 15%干酵母20 mL.kg-1;阿司匹林组:ig阿司匹林0.3 g.kg-1;给药组:分别ig MDE(1,0.5,0.25 g.kg-1),qd,预防性连续给药7 d(n=10),观察MDE对发热大鼠直肠温度的影响,测定给药后不同时间的体温变化,并采用放免法测定下丘脑中前列腺素E2(PGE2)和环磷腺苷(cAMP)含量。结果:1,0.5,0.25 g.kg-1MDE具有不同程度的退热作用;并能减少致热大鼠下丘脑中PGE2及cAMP的含量,高剂量组(126.86±24.89)ng.g-1,(9.72±1.11)nmol.g-1;中剂量组(180.12±34.58)ng.g-1,(10.05±0.26)nmol.g-1;低剂量组(203.54±30.46)ng.g-1,(10.79±0.57)nmol.g-1;与模型组(253.70±43.96)ng.g-1,(12.15±3.11)nmol.g-1比较具有显著性差异(P<0.01,P<0.05)。结论:MDE具有解热作用,其作用机制与降低下丘脑中PGE2以及cAMP有关。
Objective:To study the anti-inflammatory and antipyretic effect and mechanism of the ethanol extracts from Musca domestica larvae(MDE) on febrile rats.Method:The rats fever model were made by dry yeast.Saline group was given equal volumes saline(ig);Model group was given equal volumes saline(ig)+ sc 15% dry yeast 20 mL·kg-1 of subcutaneous injection;Treatment group: MDE(1,0.5,0.25 g·kg-1),ig qd for 7 d,Rectum temperature(ΔT/ ℃)was measured and hypothalamus proataglandin E2(PGE2) and adenosine cyclophosphate(cAMP) were detected by RIA.Result:1,0.5,0.25 g·kg-1MDE had antipyretic effect on febrile rats and could reduce the content of PGE2 and cAMP ·kg-1)(126.86±24.89) ng·g-1,(9.72±1.11) nmol·g-1],·kg-1)(180.12±34.58) ng·g-1,(10.05±0.26) nmol·g-1],·kg-1)(203.54±30.46) ng·g-1,(10.79±0.57) nmol·g-1],campared to the control(253.70±43.96) ng·g-1,(12.15±3.11) nmol·g-1)(P〈0.01,P〈0.05).Conclusion:The results indicate that ethanol extracts from M.domestica larvae has anti-inflammatory and antipyretic effect,the mechanism may be related to reducing PGE2 and cAMP.
