摘要
建立了分光光度法测定发酵液中的1,3-二羟基丙酮。以二苯胺为显色剂,考察了显色剂用量、反应温度、反应时间、显色稳定时间等因素,确定优化试验条件。本法的线性范围为0.1~0.4mg/ml,样品回收率为100.1%和102.4%。同时考察结果显示,发酵液成分和发酵液中菌体自溶物等不影响测定。
A spectrophotometry method was established for the determination of 1,3-dihydroxyacetone in the fermentation broth. Diphenylamine was used as the chromogenic reagent. Several factors, including the amount of chromogenic reagent, reaction temperature and time and color stability time, were investigated to optimize the test conditions. The calibration curve was linear in the concentration range of 0.1 - 0.4 mg/ml. The average recoveries were 100.1% and 102.4 %. The results also showed that the composition and cell autolysates in the fermentation broth had no significant influence on this method.
