3种白细胞滤器对机采血小板白细胞去除效果的对比研究 被引量:1

摘要 目的评价3种去白细胞滤器对机采血小板的滤白效果及过滤前后机采血小板质量的变化。方法随机选取60U机采血小板在(22±2)℃条件下振荡保存24~96h。分别用3种不同滤器对血小板进行过滤,检测机采血小板过滤前后的血小板计数(PLT)、血小板单位容量、白细胞量、pH值、乳酸脱氢酶(LDH)浓度、K+浓度、血小板膜表面CD62p表达率,计算白细胞残留量和血小板回收率。结果与过滤前相比,各组过滤后机采血小板pH值、LDH浓度、K+浓度和CD62p表达率均无显著差异(P>0.05);白细胞残留量达到了医药行业标准;过滤后3组间血小板膜表面CD62p表达率(均数差为2.8%、5.4%、2.6%)和血小板回收率[(92.6±6.3)%、(82.6±4.4)%、(87.4±2.7)%]有显著差异(P<0.01)。结论 3种白细胞滤器均可有效去除机采血小板中的白细胞,过滤后的机采血小板符合血液质量要求。A组滤器过滤后能保持较高的血小板回收率并有效避免血小板激活。 Objective To compare the efficiency of leukocyte and platelet depletion of three filters, and investigate the influence on the quality of apheresis platelets before and after leukocyte depletion. Methods Sixty units of donor apheresis platelets were preserved with agitation at (22±2)℃ for 24~96 h and were randomly assigned to three groups and then filtered using three types of polyester flatbed filters (A、B、C group). The platelet count, volume of apheresis platelets, leukocyte count, pH value, lactate dehydrogenase (LDH) concentration, K + concentration and CD62p expression level were detected before and after filtration. The rate of leukocyte depletion and platelet recovery were calculated. Results The results showed that the pH value, lactate dehydrogenase (LDH) concentration, K+ concentration and CD62p expression level were not significantly different between the three groups before and after filtration (P 0.05). The residual leukocytes number met the Medical Industry Standard and the CD62p expression level and platelet recovery rate among the three groups were significantly different (P 0.01). Conclusions The three filters could efficiently deplete leukocytes in the platelets and the platelets filtered meet the requirement of the blood quality. Moreover, Filter A group maintain a high platelet recovery rate and can efficiently avoid platelet activation in vitro.
