摘要
采用纳米金标记结合银染信号放大技术建立了检测福氏志贺氏菌的新方法。优化了酶标板包被亲和素使用浓度以及分子杂交和化学发光检测实验条件。结果表明,化学发光法比吸光度法检测灵敏度高15倍,通过银染信号增强后,化学发光检测目标菌DNA的检测限达2 fmol/L,相对化学发光强度与目标菌DNA浓度在5~1000 fmol/L范围内呈良好的线性关系,相对标准偏差RSD为4.2%。
A novel and rapid method by nanogold label and silver enhancement for detection of Shigella flexneri(S.Flexneri) in food was developed.Some parameters such as the concentration of avidin,and condition of molecular hybridization and chemiluminescence were optimized.Results showed that the sensitivity of chemiluminescence was 15 times higher than those of absorbance,with had low detect limit of 2 fmol/L target DNA of S.Flexneri,and a good linearity in the range of 5-1000 fmol/L.The RSD was 4.2%.
