基于流感A&B核蛋白的高灵敏度免疫层析的快速诊断方法的建立及临床应用

摘要 本试验利用高特异性人流感病毒的核蛋白特异性单克隆抗体,以双抗体夹心法原理建立同时针对流感A、流感B进行检测快速现场诊断方法,同时评价方法灵敏度,特异性等性能,并对比同类参比试剂对临床样本进行检测,比较试剂盒与市售产品的符合率。结果表明:该检测方法对流感A型毒株,B型毒株的检测灵敏度均可以TCID50达到1:1000倍稀释检出,与其他常见病毒未见交叉反应。流感A&B检测试剂盒与市售同类方法产品对比,临床检测1928例发热疑似H1N1病人样本,A型,B型流感的阳性符合率分别为95.75%和92.52%,阴性符合率分别为92.95%和99.53%。本研究产品灵敏度高,特异性好,阴阳性检出率高于市售同类产品,可以用于流感A,流感B的现场快速检测。 A double antibody sandwich fast test method was established by choosing high specific influenza A &B nucleoprotein, and its sensitivity and specific were investigated. And then fever contributed pharyngeal swabs of 1928 patients were tested by this method and the Consistency for Influenza A and B was compared with that by commercial GICA assay. The sensitivity for influenza A&B was diluted to 1:5000 for TCID50, and no cross-reaction was found with other common virus. Positive coincidence rote for influenza A were 95.75% and 92.52%, Negative coincidence rate for influenza B were 92.95% and 99.53%, respectively. This fast test method showed high sensitivity and specificity, which had the great potential in the management ofinfuluenza A&B disease on patients with fever.
